どこに is "where". どこにも is used with ない and means "nowhere" or "not anywhere".

A: I can't see any difference between the two sentences. The first sentence sounds very formal, though. We usually say, どこへ行くんですか, instead.

I just looked up the difference and found out that the first one could be used for a reproof. For example, if your senior colleague got angry for some reason in an business meeting, suddenly stood up and tried to leave the room, you would say, どこへ行くんですか. This is not a question about his destination, but a reproof of his misbehavior.
